dc.descriptionLos hidrolizados de proteínas pueden ser adicionados para mejorar las propiedades funcionales, nutricionales e inmunológicas del alimento original. Con el objeto de estudiar los grados de hidrólisis obtenidos de los concentrados de proteína de frijol Pinto Durango y de suero de leche utilizando una enzima alcalasa, se utilizó un biorreactor tipo lote (BTL) y uno de tipo membrana (BTM). Para la hidrólisis del suero de leche se compararon tratamientos consistentes en diferentes concentraciones iniciales de enzima y sustrato, así como también el uso o no de un precalentamiento a 85 °C por 15 min., fijos pH y temperatura de 8 y 50°C respectivamente, cada tratamiento se monitoreó 180 minutos. Se ajustaron las curvas de grados de hidrólisis de cada tratamiento a modelos exponenciales mediante técnicas de regresión no lineal; las correspondientes constantes cinéticas fueron estimadas. No se identificó ninguna relación con el modelo de Michaelis-Menten. El mecanismo de reacción, incluyendo un patrón de no inhibición debido al sustrato o al producto fue elucidado; una reacción de segundo orden con una inactivación enzimática fue confirmada. Para el frijol Pinto Durango se compararon tratamientos en BTL que consistían en el uso o no de precalentamiento y en BTM también la recirculación (180 minutos) o no recirculación (45 minutos), fijos pH en 8 y temperatura en 50°C. La hidrólisis se incrementó con el precalentamiento, obteniéndose casi el doble de hidrolizados comparado con los que no fueron precalentados. Se aprecia también que los hidrolizados obtenidos por BTM sin recirculación y con precalentamiento, son equiparables a BTL en los primeros 45 min. Se verificó la hidrólisis observando el perfil de los péptidos mediante una electroforesis unidimensional; se evaluó su capacidad antioxidante mediante un ensayo ABTS. En los hidrolizados del BTM se observó que los péptidos muestran menor capacidad antioxidante comparado con los obtenidos en BTL.es_ES
dc.descriptionProtein hydrolysis products can be used to improve functional, nutritional and immunological properties of food. A stirred batch reactor (BR) and a membrane reactor (MR) were used to study the hydrolysis degree of Pinto Durango bean¿s protein concentrate and whey protein concentrate using alcalase as the enzyme for the reaction. Different treatments, consisting of different initial concentrations of enzyme as well as the presence or not of a preheating treatment at 85 ° C for 15 min, were tested for whey protein hydrolysis. All treatments were carried out at pH 8 and 50 ° C and each treatment was monitored during 180 minutes. The resulting curves ofhydrolysis degree were adjusted for each treatment to exponential models using nonlinear regression techniques; the corresponding kinetics constants were estimated. A non Michaleis - Menten relationship was identified. The reaction mechanism, including a non inhibitory pattern due to substrate or product was elucidated; second order enzyme inactivation, was confirmed. In the case of the Pinto Durango beans, treatments in BR incluiding or not preheating treatment and MB recirculation (180 minutes) or not recirculation (45 minutes) were compared; all treatments were set at pH 8 and 50 ° C. The hydrolysis degree was increased by the preheating treatment, resulting in almost hydrolysis products compared to those which were not preheated. It was also observed that the hydrolysates obtained by MB without recirculation in presence of the preheating treatment, were comparable to the BR-treated during the first 45 min. The hydrolysis degree was verified by observing the peptides profile using a one-dimensional electrophoresis technique; the antioxidant activity of hydrolysates was assessed by ABTS assay. MB-hydrolysates showed peptides with lower antioxidant capacity compared with those obtained by BR.es_ES
