Description: La modificación proteica es una alternativa para generar proteínas con aplicaciones terapéuticas. En nuestro laboratorio se modificó la subunidad ácida de la amarantina con péptidos antihipertensivos (ACM3) y se expresó en la cepa Escherichia coli Rosetta (DE3). En esta investigación se sobreexpreeso la proteína ACM3 a nivel fermentador en la misma cepa mediante el control de diferentes factores: la composición del medio de cultivo, los inductores (IPTG o lactosa), el flujo de aire, la temperatura del cultivo, la agitación y la hora de la inducción. El objetivo de este trabajo fue optimizar la expresión de ACM3 en Escherichia coli Rosetta utilizando medios mínimos y lactosa como inductor. Las fermentaciones a nivel matraz mostraron que os medios mínimos A6 y A7 en combinación con el 0.5% (p/v) de lactosa permitieron la acumulación de la proteína recombinante. Entonces, para mejorar los rendimientos de ACM3 a nivel fermentador se probaron los medios A6 y A7, el inductor lactosa y diferentes condiciones fisicoquímicas. Los experimentos se realizaron mediante un diseño Plackett-Burman con puntos centrales para observar factores con efectos significativos. Los factores significativos en A6 fueron la peptona, el pH, la agitación y la inducción con lactosa. En el medio A7, la concentración de ACM3 fue afectada por la triptona, la agitación, la inducción con lactosa, el momento y la temperatura de inducción. Así, los factores se ajustaron según los resultados del análisis estadístico y los rendimientos de ACM3 se incrementaron de 303.2 a 1,531 mg/L en el medio A6 y de 363.4 a 1,681 mg/L en el medio A7. La mayor productividad se alcanzó en 4 h (~190 mg/L¿h) y 6 h (~203 mg/L¿h) de inducción en los medios A6 y A7, respectivamente. El efecto antihipertensivo de la proteína ACM3 que se expresó en los medios A6 y A7 se observó en ensayos in vitro por la inhibición de la enzima convertidora de angiotensina que mostró un IC50 de 0.043 mg/mL y 0.041 mg/mL, respectivamente. Entonces, la expresión de ACM3 en diferentes medios no generó diferencias significativas en la inhibición de la enzima convertidora de angiotensina (P¿0.05).
Protein modification is an alternative for creating proteins with therapeutic applications. In our laboratory an acid sub-unit of amaranthine with antihypertensive peptides (ACM3) was modified and expressed in the Rosetta Escherichia coli (DE3) strain. In this research the ACM3 protein at a fermenting level was over-expressed in the same strain by controlling different factors: composition of the culture, inductors (IPTG or lactose) air flow, culture temeperature, speed agitation and time induction. The objective of this work was to optimize the ACM3 expression in Rosetta Escherichia coli using minimum media and lactose as an inductor. Fermentations at flask level showed that the A6 and A7 minimum media in combination with 0.5% (w/v) lactose allowed for the accumulation of the recombinant protein. Then, to improve the yields of ACM3 at fermenting level, the A6 and A7 media, lactose inductor and different physicochemical conditions were tested. The experiments were carried out using a Plackett-Burman design with central points to observe factors with significant effects. The significant factors in A6 were peptone, the pH, speed agitation and lactose induction. In medium A7, the concentration of ACM3 was affected by tryptone, speed agitation, lactose anduction and time and temperature of induction. The factors were adjusted according to the results of the statistical analtsis, and the yields of ACM3 increased from 303.2 to 1,531 mg/L in medium A6 and from 363.4 to 1,681 mg/L in medium A7. The greatest productivity was reached in 4 h (~190 mg/L¿h) and 6 h (~203 mg/L¿h) of induction in the A6 and A7 media, respectively. The anti-hypertensive effet of the ACM3 protein expressed in the A6 and A7 media was observed in in vitro trials through the inhibition of the angiotensin converting enzyme that showed an IC50 of 0.043 mg/mL and 0.041 mg/mL, respectively. Thus the ACM3 expression in different media did not create significant differences in angiotensin converting enzyme inhibition (P¿0.05).
