dc.descriptionLa inflamación crónica tiene un papel determinante en el cáncer colorrectal. La verdolaga (Portulaca oleraceae) es un vegetal comestible con efectos farmacológicos incluyendo capacidad analgésica y anti-inflamatoria; también posee un alto contenido de fenoles y flavonoides, y su extracto metanólico posee una alta capacidad antioxidante. La inducción de enzimas de fase 2 y antioxidantes es un mecanismo efectivo por algunos fenoles y flavonoides para proteger contra carcinógenos, por lo tanto, resulta atractivo evaluar el efecto quimioprotector de la verdolaga en un modelo de inflamación-cáncer de colon inducido químicamente en ratas F344. Ratas macho fueron inducidas con 3 inyecciones subcutáneas de dimetilhidrazina (DMH, 40mg/Kg) durante una semana y, posteriormente, se les administró el dextrán sulfato de sodio (DSS, 1%) en el agua de beber como agente promotor de la inflamación de colon. Para caracterizar el modelo, los animales se sacrificaron a diferentes tiempos, observándose un efecto después de 10 semanas de tratamiento. Para seleccionar la dosis y el extracto, ratas F344 recibieron intragástricamente el extracto acuoso (EA) o metanólico (EM) de verdolaga (50, 100 y 200mg/Kg) 3 veces por semana. 24 horas después, los animales se sacrificaron y se les extrajo colon e hígado para evaluar la actividad de gutatión-S-transferasa (GST) y quinona reductasa (QR), observándose un mayor efecto inductor de estas enzimas con EA (100mg/Kg). En el modelo de inflamación-cáncer de colon, el EA (100mg/Kg) se administró intragástricamente durante 10 semanas. A pesar de que la actividad de GST en hígado y colon decreció después de 10 semanas de tratamiento con EA, la actividad hepática de QR se indujo significativamente en todos los grupos tratados; asimismo, la actividad de QR en colon se elevó con el EA (1.2-veces) en el grupo EA/DMH/DSS. En la evaluación de fosas crípticas aberrantes, el grupo EA presentó 0% de incidencia; mientras que en el grupo EA/DMH/DSS disminuyó 7.3%, con respecto al control positivo (DMH/DSS). Similarmente, los grupos tratados con EA presentaron el menor grado de inflamación, confirmando la actividad anti-inflamatoria de la verdolaga. Estos resultados indican que el extracto de verdolaga (EA, 100mg/Kg) protege contra el desarrollo de cáncer de colon asociado a inflamación en ratas.es_ES
dc.descriptionInflammation plays an important role in colorectal cancer. Purslane (Portulaca oloraceae) is an edible vegetable, with some pharmacological effects including analgesic and anti-inflammatory, and high levels of phenols and flavonoides; furthermore, its methanolic extracts exhibit high antioxidant capacity. The induction of phase 2 and antioxidant enzymes is an effective mechanism of some phenols and flavonoides to protect against carcinogens. Therefore, it is highly attractive to evaluate the chemoprotective effect of purslane on an inflammation-colon cancer model in male F344 rats. Rats received 3 s.c. injections of dimethylhydrazine (DMH, 40mg/Kg) during one week, followed by administration of dextran sodium sulfate (DSS, 1%) in drinking water to induce colon inflammation. To characterize the colon carcinogenesis-inflammation model, rats were sacrificed at different times, showing histological changes at 10 weeks of treatment. To select the dose and the extract, rats received an aqueous (AE) or methanolic (ME) extract of purslane (50, 100 and 200mg/kg, by gavage) 3 times a week. 24 hours after the last oral intubation, animals were euthanized; the liver and the colon were removed for measurement of glutathione-S-transferase (GST) and quinone reductase (QR) activities, showing the highest enzyme induction with AE (100mg/kg). In the inflammation-cancer model, AE (100mg/kg) was orally administered in rats treated with DMH/DSS for 10 weeks. Although GST activity decreased in the liver and colon after 10 weeks of AE administration, hepatic QR activity was significantly induced in all treated groups, compared to control group. In colon, QR activity was also induced with AE (1.2-fold) in DMH/DSS group. Aberrant crypt foci were also evaluated, showing a 0% incidence in the AE group, while decreasing by 7.3% in the AE/DMH/DSS group compared to positive control (DMH/DSS). Similarly, groups treated with AE presented the lowest inflammation grade compared with the control group, assuring/supporting the anti-inflammatory effect of purslane. These results confirm the chemoprotective effect of purslane extracts (AE,100mg/Kg) in an inflammation-colon cancer model in rats.es_ES
