dc.descriptionLa leucemia es el cáncer infantil más común y representa aproximadamente el 30 % de todos los cánceres en niños. Hay distintos tipos de leucemia, dependiendo de su progresividad respecto al tiempo y el linaje celular afectado; la leucemia linfoblástica aguda tipo T (LLA-T), es un tipo de leucemia que se caracteriza por su agresividad, debido a la presencia de varias alteraciones citogenéticas que determinan su resistencia a los fármacos que conforman el protocolo de quimioterapia, donde la vincristina es uno de los principales antineoplásicos que se utilizan en el tratamiento. Sin embargo, el uso de vincristina está asociado a efectos adversos como neuropatía sensorial y motora progresiva. Debido a que la LLA-T se caracteriza por la resistencia a la quimioterapia y requiere de protocolos intensivos de administración de los fármacos, las investigaciones se centran en buscar alternativas que coadyuven al tratamiento y ayuden a disminuir los efectos colaterales. Bajo este contexto, en este trabajo de investigación, se estudió el efecto antiproliferativo de betanina y su efecto coadyuvante con vincristina en células de LLA-T. Los resultados indican que la betanina disminuye la proliferación de las células de LLA-T evaluados mediante el ensayo de exclusión azul de tripano y, que al combinarse con vincristina, potencia el efecto de manera dependiente de la concentración. Además, la betanina a las concentraciones propuestas no tuvo efectos tóxicos en células mononucleares de sangre periférica, como referencia de su inocuidad en células sanas. Así mismo, la combinación de betanina y vincristina, disminuye la cantidad de células viables, aumenta la cantidad en apoptosis temprana y arresta el desarrollo de las células primordialmente en fase G2/M, determinado por citometría de flujo. Este es el primer reporte del efecto de la betanina y vincristina en células de LLA-T, así como su asociación a apoptosis y arresto del ciclo celular.es_ES
dc.descriptionLeukemia is the most common childhood cancer and accounts for approximately 30% of all cancers in children. There are different types of leukemia, depending on their progressivity with respect to time and the affected cell lineage; T-cell acute lymphoblastic leukemia (T-ALL) is a type of leukemia that is characterized by its aggressiveness, due to the presence of several cytogenetic alterations that determine its resistance to the drugs that make up the chemotherapy protocol, where vincristine is one of the main antineoplastic that are used in the treatment. However, the use of vincristine is associated with adverse effects such as progressive sensory and motor neuropathy. Because T-ALL is characterized by resistance to chemotherapy and requires intensive protocols for drug administration, research focuses on finding alternatives that contribute to treatment and help to reduce side effects. In this context, in this research work, the antiproliferative effect of betanin and its adjuvant effect with vincristine in T-ALL cells were studied. The results indicate that betanin decreases the proliferation of the T-ALL cells evaluated by the trypan blue exclusion test and, when combined with vincristine, potentiates the effect in a concentration-dependent manner. In addition, betanin at the proposed concentrations had no toxic effects on peripheral blood mononuclear cells (PBMC), as a reference for its safety in healthy cells. Likewise, the combination of betanin and vincristine decreases the number of viable cells, increases the amount in early apoptosis arrests the development of cells primarily in the G2/M phase, determined by flow cytometry. This is the first report of the effect of betanin and vincristine on T-ALL cells, as well as its association with apoptosis and arrest of the cell cycle.es_ES
