dc.descriptionEn las últimas décadas se ha estudiado el efecto de extractos de diferentes especies vegetales en diversos tipos de células incluyendo células cancerígenas, en estos estudios se ha observado un efecto citotóxico y/o antiproliferativo a nivel in vitro, los autores de dichos estudios mencionan que algunos de los compuestos bioactivos de las especies vegetales son los responsables de estos efectos aludiendo a que presentan capacidad antioxidante y diferentes compuestos antioxidantes, siendo interesante evaluar estos compuestos provenientes de otras especies vegetales de interés alimentario como lo es la hoja de Amaranto ya que a diferencia de otras partes de la planta de Amaranto, como la semilla ó el tallo, la hoja posee el mayor contenido de compuestos con capacidad antioxidante, estos pueden participar en la prevención de diversas enfermedades incluyendo cáncer, ya que pueden actuar como secuestradores biológicos de los radicales libres, el interés en los últimos años ha pasado de lo preventivo al posible tratamiento con dichos compuestos antioxidantes de manera que ciertos compuestos fenólicos sirven como moléculas señalizadoras en rutas de muerte celular. El objetivo de este trabajo fue evaluar la capacidad antioxidante y los compuestos responsables de los extractos liofilizados acuoso y metanólico de la hoja de Amaranto Amaranthus hypochondriacus l. y con los mismos determinar la citotoxicidad sobre células de cáncer de colon humano (HT-29) y fibroblastos murinos (3T3), utilizando citometría de flujo para evaluar parámetros de muerte celular. Se realizó una extracción acuosa y otra metanólica a partir de la hoja de amaranto y estos fueron rotaevaporados y liofilizados. Se evaluó la capacidad antioxidante (CA) por los métodos (FRAP y DPPH), se determinó el contenido de compuestos fenólicos totales (CFT) por el método de Folín-Cicalteau, Antocianinas (AT) por diferenciación de PH y Taninos Condensados (TC) por el método de vainillina-catequina. Se determinó la concentración inhibitoria media (CI50) y la concentración letal media (CL50) posteriormente se utilizó un kit de Anexina V y Muerte Celular para determinar apoptosis y un kit de Lactato Deshidrogenasa (LDH) para determinar necrosis en las células tratadas con los Extractos Liofilizados (EL). Se realizó un análisis de varianza (ANOVA) seguido por una prueba pos hoc de Tukey y Dunnet con un nivel de significancia (p <0,05) utilizando el programa estadístico SPSS v21 y el Software Prism GraphPad v5.es_ES
