Title: Aplicación de Elicitores AS, Quitosan y H2O2 EN Capsicum annuum y su efecto de respuesta a estrés biótico

Description: Con la finalidad de evaluar la función de los elicitores en la Resistencia Sistémica Inducida (ISR) que conlleva a la tolerancia y/o resistencia a estrés biótico en plantas, se llevó a cabo la aplicación foliar de ácido salicílico (AS), peróxido de hidrógeno (H2O2) y quitosán (QN) en plantas jóvenes de Capsicum annuum L. Para lo cual se utilizaron tres concentraciones de cada elicitor y fueron aplicadas en dos periodos de tiempo (día 0 y día 42). Donde la producción endógena de H2O2, actividades enzimáticas CAT-PAL; así como la expresión de los transcritos de cat1, pal y pr1, fueron evaluados como parte del estrés oxidativo generado por los elicitores. Así mismo, se realizó la interacción C. annuum L¿Elicitor¿Estrés biótico (PepGMV, P. capsici y X. capmpestris pv vesicatoria); y los trascritos de mkk1, mk1, mk2, wrkyd, wrky1, npr1, pr1 involucrados en la ruta de señalización intracelular MAP quinasas, fueron evaluados mediante RT-PCR. Por otro lado, se analizaron mediante HPLC los metabolitos secundarios inducidos en la interacción C. annuum L¿18 mM H2O2¿PepGMV; debido a que fue el tratamiento que mejor fenotipo de tolerancia mostró al estrés biótico. Nuestros resultados muestran que la aplicación foliar de elicitores en C. annuum L., induce la ISR. Lo cual pudo comprobarse con el alto contenido endógeno de H2O2 (5 ng/mg PF) generado con 6.7 mM AS y 18 mM H2O2, respecto al control con agua. Mientras que los tratamientos con QN, indujeron las mismas respuestas en menor proporción que los otros dos inductores. Así mismo, en las interacciones planta¿elicitor¿patógeno, se identificaron fenotipos tolerantes con los tratamientos 0.1 mM AS, 18 mM H2O2 y 1000 ¿g/ ¿L QN. De igual forma, se confirmó que la aplicación foliar de elicitores, induce la expresión de los genes involucrados en la ruta principal de señalización intracelular, MAP quinasa. Así como la producción de metabolitos secundarios como ácido cafeico, ácido elágico, rutina, ácido rosmarínico, quercetina y resveratrol, los cuales están involucrados en el sistema de defensa de las plantas.
In order to evaluate the function of the elicitors on Induced Systemic Resistance (ISR) leading to tolerance and / or resistance to abiotic stress in plants, was conducted foliar application of salicylic acid (SA), hydrogen peroxide (H2O2) and chitosan (QN) in young plants of Capsicum annuum L. for which three concentrations of each elicitor were used and were applied in two time periods (day 0 and day 42). Where the endogenous production of H2O2, CAT-PAL enzyme activities; and the expression of transcripts of cat1, pal and pr1 were evaluated as part of the oxidative stress generated by elicitors. Likewise, the interaction C. annuum L-Elicitor-biotic stress (PepGMV, P. capsici and X. pv vesicatoria capmpestris) was performed; and transcripts of mkk1, mk1, mk2, wrkyd, wrky1, npr1, pr1 involved in intracellular signaling pathway MAP kinases were assessed by RT-PCR. On the other hand, secondary metabolites induced by C. annuum L-18 mM H2O2-pathogen interaction, was analyzed by HPLC because it was the treatment that best phenotype showed tolerance to biotic stress. Our results show that foliar application of elicitors in C. annuum L., induces ISR. Which, could be verified with the high endogenous content of H2O2 (5 ng / mg WF) AS generated with 6.7 mM H2O2 and 18 mM with respect to the water control. While, QN treatments, induced the same responses but a lesser extent than the other two inductors. Also, in the plant-elicitor-pathogen interactions were identified treatments 0.1 mM SA, 18 mM H2O2 and 1000 ug / uL QN. Similarly, it was confirmed that the foliar application of elicitors, induces expression of genes involved in intracellular signaling pathway leading, MAP kinase. Just as the production of secondary metabolites such as caffeic acid, ellagic acid, rutin, rosmarinic acid, quercetin and resveratrol, which are involved in the defense system of plants.
