Descripción : Los adipocitos son las células principales del tejido adiposo. Una de sus funciones es el almacén de energía en forma de triglicéridos. Existen tres tipos de adipocitos diferenciados por sus características y marcadores particulares, el blanco, marrón y beige, de los cuales, los dos últimos tienen la capacidad de liberar energía en forma de calor debido a un proceso llamado termogénesis. Como alternativa a la patología de obesidad se buscan factores importantes que promuevan el aumento en número y actividad de estos últimos, por su capacidad termogénica, siendo de mayor relevancia el beige por compartir depósitos con el tejido adiposo blanco. Estudios previos han mostrado los diversos efectos de la hormona de crecimiento (somatotropina) en el tejido adiposo como: la lipólisis, la diferenciación de preadipocitos y la expresión de la proteína UCP-1 para aumentar la termogénesis en el tejido adiposo marrón. Por lo tanto, el objetivo del presente trabajo fue determinar el efecto de la hormona de crecimiento en la diferenciación de preadipocitos a adipocitos beige mediante la determinación de uno de los marcadores principales de los adipocitos beige, CD137. Se trabajó con tres grupos de células 3T3-L1: uno con inducción a adipocito blanco y otro con inducción a adipocito beige, como controles, y un tercer grupo con inducción a adipocito beige suplementado con hormona de crecimiento. Se observaron los cambios en la forma y complejidad de las células mediante microscopía de luz a lo largo de todo el proceso al igual que el grado de diferenciación mediante la técnica de rojo oleoso. De igual forma se evaluó la expresión proteica de CD137 mediante citometría de flujo, al igual que los marcadores CITED1, en adipocitos beige, y TCF21, en adipocitos blancos, mediante western blot y PCR punto final. Los resultados mostraron que no existe diferencia estadística significativa en la acumulación de lípidos entre los grupos de células diferenciadas, sin embargo, si existe diferencia en la forma de las gotas lipídicas así como su organización en la célula. De igual forma, se corrobora la diferenciación a adipocito blanco con la expresión de TCF21 y la diferenciación a adipocito beige con la expresión de CITED1, sin embargo, al comparar entre todos los grupos mediante citometría de flujo para el marcador CD137, el grupo de células con el estímulo de diferenciación a adipocitos beige con y sin hormona de crecimiento presentaron menor expresión del marcador respecto a los demás grupos, contrario a lo esperado. De acuerdo a los resultados, y bajo las condiciones de exposición y tratamiento, la hormona de crecimiento no tiene efecto sobre la expresión del marcador CD137 en el proceso de diferenciación a adipocito beige.
Adipocytes are the main cells of adipose tissue, having as one of its main functions the storage of energy in the form of triglycerides. There are three types of adipocytes differentiated by their characteristics: white, brown and beige. The brown and beige adipocytes have the ability to release energy as heat through a process called thermogenesis. As an alternative to the pathology of obesity, it is important to develop factors that promote the increase in number and activity of the brown and beige adipocytes, being more important for us, the adipocyte beige because it shares deposits with white adipose tissue. Previous studies have shown the different effects of growth hormone (somatotropin) in adipose tissue as lipolysis, preadipocyte differentiation and expression of UCP-1 protein to increase thermogenesis in brown adipose tissue. Therefore, the objective of this work was to determine the effect of growth hormone in the differentiation of preadipocytes cell line 3T3-L1 to beige adipocytes by the measure of one of the markers of beige adipocytes, CD137. We used three groups of cell culture, one was worked with white induction and the second with beige induction. The third group was the beige induction with growth hormone. The adipocyte morphology and differentiation was determined by the oil red procedure and light microscope observation. Finally, the protein expression of CD137 was evaluated as one of the specific markers of beige adipocytes. We evaluated changes in the shape and complexity of the cells by light microscopy throughout the whole process as well as the degree of differentiation by the oily red technique. In the same way, CD137 protein expression was evaluated by flow cytometry, as well as CITED1 marker in beige adipocytes and TCF21 in white adipocytes by western blot and end point PCR. The results showed that there is no significant statistical difference in lipid accumulation between groups of differentiated cells, however, we showed differences in the form of lipid droplets as well as their organization in the cell. We corroborated the differentiation to white adipocyte with the expression of TCF21 and the differentiation to adipocyte beige with the expression of CITED1. However, when comparing all groups by flow cytometry for the marker CD137, the group of cells with the stimulus of differentiation to beige adipocytes with and without growth hormone showed lower expression of the marker with respect to the other groups, contrary to what was expected. According to the results, and under the conditions of exposure and treatment, the growth hormone has no effect on the expression of the marker CD137 in the process of differentiation to beige adipocyte.
