Description: Los extractos de plantas del género Crataegus son actualmente el principal tratamiento alternativo de las enfermedades cardiovasculares en Europa, lo que se ha justificado con diversas investigaciones en las que se demostraron sus efectos vasodilatadores, hipotensivos y cardioprotectores. Estos efectos se atribuyen a la gran variedad de constituyentes químicos que se puede encontrar en estos extractos y cuya caracterización podría permitir el establecimiento de los mecanismos mediante los cuales actúan en el organismo, así como la estandarización de su composición química con el fin de asegurar su efecto terapéutico. Las evidencias de los efectos favorables de distintas especies de Crataegus sobre el sistema cardiovascular son un buen indicador del potencial de la especie mexicana Crataegus gracilior (Rosaceae) como agente medicinal y como una fuente de compuestos farmacológicamente activos, por lo que en este estudio se realizó el análisis de los constituyentes químicos de una fracción del extracto metanólico de esta especie, que presentó un efecto vasorrelajante eficaz en una fase previa de este proyecto. El análisis consistió en la aplicación de distintas técnicas de separación, principalmente las cromatografías en columna y capa fina, con la finalidad de disminuir la complejidad de la muestra. Posteriormente, se purificaron algunos compuestos haciendo uso de la cromatografía de líquidos de alta resolución. Finalmente, la espectroscopia de resonancia magnética nuclear en sus diferentes modalidades permitió la elucidación completa de las estructuras químicas de tres moléculas: un nor-triterpeno monoglicosilado y dos flavanoles, uno de ellos sustituido por una cumarina. Conviene destacar que éste es el primer reporte de la presencia de este derivado cumarínico en el género Crataegus y el segundo en el reino vegetal, ya que sólo ha sido descrito en Anthocephalus cadamba (Rubiaceae).
Nowadays, extracts from plants of the genus Crataegus are the main alternative treatment of cardiovascular disease in Europe. This application has been supported by several research works through which the vasodilator, hypotensive and cardioprotective effects of these extracts have been demonstrated. These effects have been attributed to the great diversity of chemical constituents present in the extracts and whose characterization may promote the elucidation of the mechanisms by which the extracts exert their activity on the human body, and could also lead to the optimization of their efficacy and safety of their therapeutic use. The up to now proven evidence about the beneficial effects that different extracts from Crataegus species have on the cardiovascular system is an excellent indicator of the medicinal potential of Crataegus gracilior (Rosaceae), but also of its importance as a source of pharmacologically active compounds. Based on this fact, a study of the chemical constituents of a vasoactive chromatographic fraction from the methanolic extract obtained from the leaves of the Mexican species C.gracilior was undertaken. Different separation techniques, including column and thin layer chromatographies were employed in order to obtain samples of less chemical complexity. Afterwards, the individual constituents in these samples were purified by high-performance liquid chromatography. Different nuclear magnetic resonance spectroscopy techniques were used to establish the complete chemical structures of the three main purified molecules, a monoglycosylated nor-triterpene and two flavanols, one of which is substituted by a coumarin. It is worth mentioning that this is the first report of the presence of this coumarin derivative in this plant genus and the second in the plant kingdom, since it has only been described in Anthocephalus cadamba (Rubiaceae).
