dc.descriptionLas hojas de los encinos (Quercus, sp) son tradicionalmente usadas en México como alimento, y algunas infusiones tienen efectos anticancerígenos; sin embargo, esta propiedad aún no se ha comprobado. El objetivo fue estudiar el efecto anticancerígeno del consumo de infusiones de hojas de encino Q. sideroxyla y Q. durifolia en un modelo de cáncer de colon. Se prepararon infusiones de Q. sideroxyla y Q. durifolia y se determinó el contenido total de fenoles y flavonoides, y la capacidad antioxidante. La infusión de Q. sideroxyla mostró la mayor capacidad antioxidante (755.56±29 ¿mol Trolox/g de hoja seca) y la mayor concentración de fenoles y flavonoides (35.15±0.0 mg de ácido gálico y 22.34±0.3 mg de (+)-catequina/g de hoja seca) comparado con Q. durifolia. Por HPLC, se identificaron en ambas infusiones tres de las principales catequinas reportadas para el té verde (epigalocatequin_galato, epicatequina y catequina). Posteriormente, las infusiones se administraron como única fuente de bebida a ratas macho Sprague-Dawley, en concentración de 1% (p/v) durante todo el periodo experimental. En la semana 4, las ratas recibieron 8 inyecciones s.c. de 1,2-dimetilhidrazina (21mg/kg de peso corporal) una vez por la semana. Después de 26 semanas, los animales fueron sacrificados y el colon fue removido para estudios histopatológicos y moleculares. Tanto la incidencia (31 y 73%) como el promedio de tumores (adenocarcinomas, 0.8±0.41 y 2.0±0.55) fueron significativamente más bajos en el grupo tratado con Q. sideroxyla comparado con el grupo 1,2-DMH, respectivamente (p<0.05, por la prueba de ¿2), mientras que, la infusión de Q. durifolia no tuvo efecto protector en estos parámetros. Sin embrago, se observaron efectos adversos en el intestino delgado por el tratamiento con ambas infusiones. Por otro lado, la expresión de la proteína beta-catenina disminuyó significativamente (2.3 veces) en adenocarcinomas del grupo Q. sideroxyla comparado con los niveles encontrados en el grupo 1,2-DMH (p<0.05, por la prueba de Dunnet). Los resultados sugieren que el consumo de infusiones de hojas de Q. sideroxyla posee efecto anticancerígeno contra el desarrollo de cáncer de colon; sin embargo, estudios toxicológicos posteriores son necesarios para descartar cualquier efecto adverso causado por el consumo regular de esta infusión.es_ES
dc.descriptionOak leaves (Quercus sp.) are traditionally used as food in Mexico, and some infusions have anticancer effect; however, this property has been scientifically tested not yet. The aim was to study the anticancer effect of infusions of edible oak species Q. sideroxyla and Q. durifolia in a colon cancer model. Leaf infusions of Q. sideroxyla and Q. durifolia were prepared and the total phenol and flavonoid contents, and total antioxidant capacity were determined. Q. sideroxyla infusion showed the highest antioxidant capacity (755.56±29 ¿mol Trolox/g of dried leaf) and contained the highest phenol and flavonoide concentrations (35.15±0.0 mg of gallic acid and 22.34±0.3 mg de (+)-catechin/g dried leaf, respectively) compared to Q. durifolia. By HPLC analysis, three of the major catechins reported for green tea (epigallocatechin_gallate, epicatechin and catechin) were identified in both infusions. Later, these infusions were administered as the sole source of liquid to male Sprague-Dawley rats, at a 1% concentration (w/v) for the entire experimental period. On week 4, rats received 8 s.c. injections of 1,2-dimethylhydrazine (21 mg/kg body weight) once a week. After 26 weeks, animals were sacrificed and colon was removed for histopathological and molecular studies. Both the incidence (31 vs 73%) and mean tumor (adenocarcinomas, 0.8±0.41 vs 2.0±0.55) were significantly lower in Q. sideroxyla¿treated group compared with 1,2-DMH group, respectively (p<0.05, by the ¿2 test). On the other hand, Q. durifolia infusion did not show protective effect on these parameters. However, adverse effects by both infusions were observed in the small intestine of treated rats. On the other hand, the expression level of beta-catenin protein was also significantly reduced by 2.2-fold in adenocarcinomas of the Q. sideroxyla-treated group compared with level found in the 1,2-DMH group (p<0.05, by the Dunnet¿s test). Overall results suggest that the consumption of the leaf infusion of Q. sideroxyla had an anticancer effect against the development of colon cancer; however, further toxicological studies are required in order to rule out any adverse effect caused by the regular consumption of this infusion.es_ES
