Description: El ácido giberélico (GA ) es el principal metabolito secundario producido por fermentación del hongo Gibberella fujikuroi. Esta fitohormona tiene gran importancia en la agricultura e industria cervecera. En 3 esta investigación, una mezcla glucosa-aceite de maíz fue usada en tres diferentes proporciones sobre la base de 40 g L-1 de carbono en un biorreactor de tanque agitado de capacidad 7 L para la producción de GA3 por G. fujikuroi. A través del tiempo de cultivo, fueron mantenidos y controlados los valores de pH 3.5, temperatura 29 °C, agitación 600 rpm y aireación 1 vvm. El metabolismo secundario inició entre las 48 y 60 horas de cultivo y la máxima producción de GA3 se encontró a las 288 h para los tres tratamientos propuestos con fuente de carbono mixta. Se cuantificó por Cromatografía Líquida de Alta Resolución (HPLC) una producción de 430 mg L-1 de GA3 cuando se utilizó la proporción 1:2 glucosa-aceite de maíz; 380 mg L-1 de GA3 cuando se ensayó la proporción 1:1 glucosa-aceite de maíz; en contraste a los 280 mg L-1 cuando se utilizó la proporción que contenía la mayor cantidad de glucosa (proporción 2:1). Por otro lado, se obtuvieron 358 mg L-1 cuando se utilizó aceite como única fuente de carbono; y al emplear solo glucosa, el metabolismo secundario inició a las 96 h de cultivo obteniendo la máxima concentración de 136 mg L-1 de GA3 a las 264 h de fermentación, siendo así, la menor cantidad de GA3 producida durante todas las fermentaciones realizadas. Se concluyó que la fuente de carbono afecta el tiempo de fermentación así como la producción del metabolito. G. fujikuroi mostró gran capacidad para adaptarse a medios de cultivo con aceite de maíz como fuente de carbono, realizando una rápida metabolización de los ácidos grasos (los cuales fueron cuantificados por Cromatografía de Gases). Por tanto, la glucosa no ejerce ningún efecto de represión catabólica sobre la asimilación del aceite de maíz, o alguna clase de control metabólico sobre el transporte de los ácidos grasos en las células de G. fujikuroi cuando se utiliza fuente de carbono mixta.
Gibberellic acid (GA3) is the chief secondary metabolite produced by fermentation of the Gibberella fujikuroi fungus. This phytohormone is of great importance in agriculture and in the brewing industry. In this research, a glucose-corn oil mixture was used in three different proportions on the basis of 40 g L-1 of carbon in a 7 L stirred tank bioreactor for the production of GA3 by G. fujikuroi. Throughout the culture time, the values of pH 3.5, 29 °C temperature, 600 rpm agitation and 1 vvm aeration were maintained and controlled. The secondary metabolism began between 48 and 60 hours of culture, and the maximum production of GA was found at 288 h for the three treatments proposed with a mixed carbon source. A production of 430 mg of GA L was quantified by High Performance Liquid Chromatography (HPLC) when using the glucose-corn oil ratio 1:2; 380 mg GA L when the glucose-corn oil ratio was 1:1; in contrast to 280 mg L when the proportion that contained the largest amount of glucose (ratio 2:1) was used. We obtained 358 mg L when oil was used as the sole carbon source; when only glucose was used, the secondary metabolism began at 96 h of culture, obtaining the highest concentration of 136 mg GA L at 264 h of fermentation, this being the least amount of GA produced during all fermentations performed. Our conclusion is that the carbon source affected the fermentation time as well as the production of the metabolite. G. fujikuroi showed a great ability to adapt to culture media with corn oil as a carbon source, resulting in a rapid metabolism of fatty acids (which were quantified by Gas Chromatography). Therefore, glucose does not exert any catabolic repression on the assimilation of corn oil, or any kind of metabolic control on the transport of fatty acids in the cells of G. fujikuroi when using a mixed carbon source.
