Título : Efecto modulador de los frutos de arándano (Vaccinium myrtillus) y zarzamora (Rubis fructicosis) sobre biomarcadores de estrés oxidativo en ratas Wistar diabéticas inducidas con estreptozotocina

Descripción : La diabetes mellitus se caracteriza por el incremento de la producción de radicales libres y especies reactivas del oxígeno, mientras disminuye la capacidad del sistema de defensa antioxidante. Por lo que la identificación de diferentes antioxidantes en plantas y sus frutos, perece una estrategia altamente atractiva. Frutos como el arándano y la zarzamora son ricos en compuestos fenólicos, con una alta capacidad antioxidante. Para evaluar el efecto modulador de los frutos de arándano y zarzamora sobre biomarcadores de estrés oxidativo en ratas Wistar diabéticas inducidas con estreptozotocina, los frutos fueron liofilizados y se les determinó el contenido de fenoles y flavonoides totales, siendo la zarzamora con mayor contenido de fenoles y flavonoides, y mayor capacidad antioxidante (medidos por los métodos de ABTS ¿+ y FRAP). En un estudio agudo (7 días), los frutos fueron administrados intragástricamente (50, 100 y 200 mg/kg), mostrando un efecto hipoglucemiante con zarzamora (100 y 200 mg/kg) en ratas sanas y diabéticas. La actividad enzimática de glutatión-S-transferesasa (GST) y los niveles de glutatión reducido (GSH) fueron estadísticamente mayores en el hígado y riñón de ratas sanas, siendo más significativa para las ratas tratadas con zarzamora a 200 mg/kg. Los grupos de ratas diabéticas consumieron más alimento (25%) y agua (70%) que las ratas sanas, confirmando la presencia de la enfermedad. Durante la fase experimental subcrónica (21 días) se observaron incrementos significativos en los niveles de glucosa, consumo de alimento y agua, así como pérdida de peso en los grupos de ratas diabéticas tratadas con y sin zarzamora. La actividad de GST, superóxido dismutasa y catalasa, así como los niveles hepáticos y renales de GSH se encuentran disminuidos en ratas diabéticas comparadas con los de ratas sanas. Sin embargo, se observó la inducción de enzimas antioxidantes por el sulfurofano (500 ¿mol/kg) y la zarzamora (100 y 400 mg/kg) incorporada en la dieta en ratas sanas y diabéticas. El presente estudio muestra el incremento de las enzimas de defensa antioxidante en la protección contra el estrés oxidativo en pacientes diabéticos por estos frutos.
Diabetes mellitus is characterized by the increase in the production of free radicals and reactive oxygen species, while the capacity antioxidant defense system is diminished. In this regard the identification of different antioxidants in plants and their fruits, seems a highly attractive strategy. Fruits like blueberry and blackberry are rich in phenolic compounds, with a high antioxidant capacity. To evaluate the effect of blueberry and blackberry fruits on oxidative stress biomarkers in diabetic Wistar rats induced with streptozotocin, fruits were lyophilized and the content of total phenols and flavonoids was determined, being the blackberry fruit with the highest content of phenols and flavonoides, and greater antioxidant capacity (measured by ABTS ¿+ and FRAP methods) than the blueberry. In a acute study (7 days), the fruits were intragastrically administrated (50, 100 and 200 mg/kg), showing a hypoglycemic effect with blackberry (100 and 200 mg/kg) in diabetic and healthy rats. The enzymatic activity of glutatión-S-transferesasa (GST) and the levels of reduced glutatión (GSH) were statistically greater in the liver and kidney of healthy rats, being more significant for the rats with blackberry (200 mg/kg). Diabetic rat groups consumed more food (25%) and water (70%) that healthy rats, confirming the presence of the illness. During a subchronic experimental phase (21 days) significant increments in the levels of glucose, consumption of food and water, as well as loss of weight were observed in diabetic rats treated with and without blackberry. The activity of GST, superoxide dismutase and catalase, as well as the renal and hepatic levels of GSH were found diminished in diabetic rats compared with those of healthy rats. Nevertheless, the antioxidant enzyme induction for sulfurophane (500 ¿mol/kg) and blackberry fruit (100 and 400 mg/kg) incorporated into the diet was observed in diabetic and healthy rats. The present study shows the increase of antioxidant defense enzymes provided by these fruits which protect against oxidative stress in diabetic patients.
