Título : Modos de acción de agentes de biocontrol y mecanismos de resistencia de genotipos de manzana a Penicillium expansum

Descripción : El manzano es una de las especies frutícolas de mayor importancia a nivel mundial, pero su fruto se ve afectado durante su almacenamiento por la pudrición azul causada por Penicillium expansum Link, por lo que para su control se usan fungicidas de síntesis que resultan tóxicos para el ambiente y el humano. Por ello, en la actualidad se buscan alternativas como el uso de cepas antagónicas o de cultivares tolerantes al patógeno. Los objetivos de esta tesis fueron: i) caracterizar la resistencia a P. expansum de genotipos de manzana establecidos en Querétaro, México y; ii) Evaluar la efectividad y modos de acción de cepas de levaduras autóctonas contra este patógeno. Diversos genotipos de manzana se inocularon con P. expansum, se midió el desarrollo del hongo y se determinó la correlación de algunas características físicas y químicas de los frutos con la severidad del patógeno; los frutos menos susceptibles al hongo fueron sometidos a estudios de expresión de genes por PCR cuantitativo. Por otro lado, las levaduras con potencial como agentes de biocontrol se sometieron a pruebas de antibiosis en cajas de Petri y de competencia por nutrientes en placas de cultivo con filtros semipermeables. Los resultados mostraron que la susceptibilidad de los genotipos de manzana a P. expansum se vio fuertemente influenciada por el año de cosecha (2011 y 2012). En 2012 (año en que los genotipos mostraron menor susceptibilidad), los genotipos menos susceptibles fueron `436¿ y `Lourdes¿ (17.4 y 18.5 mm de diámetro de podredumbre, respectivamente) en comparación contrastando con `Golden Delicious¿ (37.9 mm) considerada como genotipo de referencia. Asimismo el estado de madurez de los frutos, más no su época de maduración, influyó en el desarrollo del hongo, siendo los frutos menos maduros los que tuvieron menor infección. Por otro lado, a excepción del contenido de sólidos solubles totales en 2012, ninguna de las variables físicas y químicas evaluadas correlacionó con la susceptibilidad de las manzanas a P. expansum. Sin embargo, los genotipos `Lourdes¿ y `436¿ mostraron cambios notables en la expresión de genes relacionados con la ruta de fenilpropanoides, proteínas PR y síntesis de etileno. Con relación a los agentes de biocontrol, se obtuvieron cepas que redujeron considerablemente el desarrollo del hongo en área (de 52.4 a 98.1 %) como 2R, M18/3B, M20/2A, B39, B38, B33 y B34. Finalmente, no se observó producción de antibióticos por ninguno de los antagonistas mencionados, pero seis de ellos inhibieron al hongo estando en contacto directo o no con éste, sugiriendo un modo de acción de competencia por nutrientes con el patógeno. Sin embargo, la cepa B38 disminuye significativamente su efecto sobre el hongo cuando las células se encuentran separadas por el filtro lo que sugiere una modo de acción de competencia por nutrientes y/o interacción directa. Las cepas M18/3B, M20/2A, y B34 fueron identificadas como Yarrowia lipolytica y la B33 como Debaryomyces hansenii.
Apple tree is one of the most important fruit species worldwide, but its fruit is affected during storage by blue rot caused by Penicillium expansum Link. In order to control it fungicides that are toxic for environment and human are commonly used. Therefore, new alternatives are searched as the use of antagonistic strains or tolerant cultivars to fungus. The objectives of this thesis were: i) characterize the resistance to P. expansum of apple genotypes established in Querétaro, Mexico and; ii) evaluate the effectivity and modes of action of native yeast strains against this pathogen. Several genotypes of apple were inoculated with P. expansum; fungal growth was measured and physical and chemical characteristics of fruit were correlated with the severity of the pathogen; least susceptible fruits were subjected to gene expression studies by quantitative PCR. Moreover, yeasts with potential as biocontrol agents were tested for antibiosis in Petri dishes and for competition for nutrients in culture plates with a semipermeable filter. The susceptibility of apple genotypes to P. expansum was strongly influenced by the harvest year (2011 and 2012). In 2012 (year where genotypes showed less susceptibility), the least susceptible genotypes were '436' and 'Lourdes' (17.4 and 18.5 mm of rot diameter, respectively) contrasting with 'Golden Delicious' (37.9 mm) considered as reference genotype. Also, the fruit ripeness, but not its ripening time, influenced the development of the fungus being the fruit less mature the least infected. Furthermore, excluding for the content of total soluble solids in 2012, neither of the physical and chemical parameters evaluated were correlated with the susceptibility of apples to P. expansum. However, genotypes 'Lourdes' and '436' showed significant changes in gene expression related to phenylpropanoid metabolism, PR proteins and ethylene synthesis. Regarding biocontrol agents, strains that reduced considerably the fungi growth in area (from 52.4 to 98.1%) were 2R, M18/3B, M20/2A, B39, B38, B33 y B34. Finally, antibiotic production was not observed by any of these antagonists, but six of them inhibited fungus being or not in direct contact with the fungus, suggesting a mode of action by competition for nutrients with the pathogen. However strain B38 reduces the inhibition of fungus when cells are separated by filter suggesting a mode of action by competition for nutrients and/or direct interaction. Strains M18/3B, M20/2A, and B34 were identified as Yarrowia lipolytica and B33 as Debaryomyces hansenii.
