Description: Las zarzamoras (Rubus spp) son frutos ricos en compuestos nutracéuticos (flavonoides, ácidos fenólicos, taninos y otros), reconocidos por su beneficio a la salud. El objetivo de esta investigación consistió en determinar el potencial alimentario y nutracéutico de una colección de zarzamoras silvestres y mejoradas de las regiones boscosas de Michoacán y compararlo con materiales comerciales. Como materiales de estudios se utilizaron seis genotipos de zarzamoras: cuatro silvestres (ZS-3, ZS-7, ZS-10 y ZS-11), una comercial ¿Tupy¿ y una línea mejorada (UM-601). A través de columnas empacadas con resinas Amberlite XAD-7 y Sephadex LH-20 se obtuvieron extractos y fracciones ricos en polifenoles. Análisis con HPLC y LC-ESI-MS revelaron que la principal antocianina en zarzamora mexicana fue cianidina-3- O-glucósido. También se identificaron proantocianidinas en forma de monómeros a hexámeros y elagitaninos. La concentración media de antocianinas para el genotipo comercial fue de 49.2 mg/g de fruto liofilizado (equivalentes de Cy-3-O-glu), las fracciones de genotipos silvestres y la línea mejorada tuvieron un intervalo de 361.3¿494.9 mg/g de fruto liofilizado (equivalentes de Cy-3-O-glu). La concentración de proantocianidinas varió en los genotipos silvestres (417.5¿1343.6 mg/g de fruto liofilizado, equivalentes de catequina). Asimismo, a los extractos obtenidos se les evaluó fenoles totales, capacidad antioxidante, potencial antimutagénico e inhibición de marcadores proinflamatorios (ON, iNOS, PGE-2 y COX-2) en macrófagos RAW 264.7. Las fracciones de proantocianidinas a una concentración de 100 ¿g/mL inhibieron fuertemente la mutagenicidad inducida por 1-nitropireno. Con 50 ¿M (equivalentes de Cy-3-glu o catequina) algunas fracciones inhibieron (p<0.05) todos los marcadores. La fracción rica en antocianinas ZS-10 y la fracción rica en proantocianidinas UM-601 fueron los inhibidores más potentes de ON (IC50=16.1 y 15.1 ¿M, respectivamente). Las fracciones ricas en proantocianidinas ZS-7 y mejorada mostraron la mayor inhibición en la expresión de iNOS (IC50=8.3 ¿M). Las fracciones ricas en polifenoles ZS-7 y UM-601 fueron los inhibidores más potentes en expresión de COX-2 (IC50=19.1 y 19.3 ¿M equivalentes de Cy-3-glu, respectivamente). Este estudio permitió demostrar que la utilización de resinas Amberlite XAD-7 y Sephadex-LH-20 permite separar y caracterizar los polifenoles de zarzamora; además, se podrían sugerir a los genotipos de zarzamora silvestre mexicana, como fuente rica en nutracéuticos, para ser utilizados en programas de fitomejoramiento para el desarrollo de nuevas variedades.
Blackberries (Rubus spp) are fruits rich in nutraceutical compounds (flavonoids, tannins, etc.) and are recognized for their health benefits. The objective of this research was to determine the nutritional and nutraceutical potential of a collection of wild and improved blackberries from the forest regions of Michoacán and compared it with commercial materials. Six genotypes of blackberries were studied: wild (ZS-3, ZS-7, ZS-10 and ZS-11), commercial ¿Tupy¿ and an improved line (UM-601). Using columns packed with Amberlite XAD-7 and Sephadex LH-20 resins, extracts and fractions rich in polyphenols were obtained. Analyses with HPLC and LC-ESI-MS showed that the principal anthocyanin in Mexican blackberries is cyaniding-3-O-glucoside. Proanthocyanidins were identified in the form of monomers to hexamers and ellagitannins. The average concentration of anthocyanins for commercial cultivar was 49.2 mg/g of lyophilized fruit; the wild and improved genotype fractions had a range of 361.3-494.9 mg/g of lyophilized fruit (equivalents of Cy-3-O-glu). The concentration of proanthocyanidins varied in the wild genotypes (417.5-1343.6 mg/g catechin equivalents). The extracts obtained were evaluated for total phenols, antioxidant capacity, antimutagenic potential and inhibition of proinflammatory markers (ON, iNOS, PGE-2 and COX-2) in RAW 264.7 macrophages. Fractions of proanthocyanidins at a concentration of 100¿g/mL strongly inhibited the mutagenicity induced by 1-nitropyrene. With 50 ¿M (equivalents of Cy-3-glu or catechin), some fractions inhibited (p <0.05) all markers. The fraction rich in anthocyanins ZS-10 and the fraction rich in proanthocyanidins UM-601 were the most potent inhibitors of ON (IC50=16.1 and 15.1 ¿M respectively). The fractions rich in proanthocyanidins ZS-7 and improved cultivar showed the highest inhibition in the iNOS expression (IC50=8.3 ¿M). The fractions rich in polyphenols ZS-7 and UM-601 were the most potent inhibitors in the expression of COX-2 (IC50=19.1 and 19.3 ¿M equivalents of Cy-3-glu, respectively). This study shows that the use of Amberlite XAD-7 and Sephadex LH-20 resins can make possible the separation and characterization of blackberry polyphenols. It can also be suggested that the genotypes of wild Mexican blackberries, as a rich source of nutraceuticals, can be used in phytoimprovement programs for the development of new varieties.
