Description: Las lectinas de leguminosas son proteínas con la capacidad de reconocer carbohidratos que se encuentren integrados a membrana o que estén de manera libre. Son consideradas factores antinutricios porque afectan la biodisponibilidad de nutrimentos. La fracción concentrada de lectinas (FCL) del frijol Tépari (Phaseolus acutifolius) presenta capacidad citotóxica selectiva en líneas celulares de cáncer, así como la capacidad para inhibir tumores de colon en etapa temprana en ratas. En estudios in vivo nuestro grupo de trabajo ha determinado que presenta buena tolerabilidad, sin afectación sistémica, sin embargo, se presentaron efectos antinutricios como la pérdida en la ganancia de peso del 10% en las ratas tratadas. Lo anterior hace necesario el estudio sobre sus efectos sistémicos y remanentes en ratas de diferentes edades, considerándose importante en el desarrollo de un fármaco. Se utilizaron ratas Sprague Dawley sanas adquiridas de la INB-UNAM de 5 y 25 semanas de edad. Los esquemas de tratamiento fueron: estudios a corto plazo (6 semanas de tratamiento) tanto en ratas adultas como jóvenes sanas; estudio intermitente (2 períodos de 6 semanas de tratamiento y 2 semanas intermedias de descanso) con ratas jóvenes en donde se realizaron sacrificios los días 1, 9, 22, 43, 57, y 100 para observar efectos remanentes y sistémicos; estudio sostenido (12 semanas de tratamiento) con ratas jóvenes. Los animales se pesaron y midieron semanalmente y se determinó el consumo de alimento. Al finalizar el tratamiento correspondiente se realizaron pruebas bioquímicas como biometrías hemáticas, proteínas totales, albúmina, creatinina, urea, α-amilasa, insulina, ALT y glucosa. El hígado, páncreas, intestino delgado y grueso, timo y bazo se pesaron, se midieron y se realizaron su análisis histopatológico. No se encontraron efectos sistémicos y antinutricios tras la administración subcrónica de la FCL con ratas adultas y jóvenes, sin embargo, se observó hipertrofia pancreática. En el experimento intermitente en ratas jóvenes se observó disminución en la ganancia de peso del 4%, al día 9 se presentó aumento en la proporción de granulocitos e incremento de la pulpa blanca en bazo, el día 43 se observó incremento de tamaño de intestino delgado del 14% y disminución del intestino grueso del 21%, atrofia intestinal, así como hipertrofia pancreática. Tras el descanso de 2 semanas, se revirtió la hipertrofia provocando atrofia pancreática y al día 100 se encontró que el páncreas se recuperó. En el tratamiento crónico no presentaron efectos sistémicos ni antinutricios, excepto el incremento del tamaño del corazón. Los resultados muestran que la administración de la FCL provoca efectos adversos relacionados con sus propiedades antinutricias sin efecto detrimental a nivel sistémico. Será necesario determinar sí los efectos adversos se revierten por completo utilizando esquemas más largos de descanso.
Legume lectins are proteins with the capacity to recognize membrane-integrated or free carbohydrates. They are considered antinutritional factors because they affect the bioavailability of nutrients. The Tepary bean (Phaseolus acutifolius) lectin fraction (TBFL) has selective cytotoxic capacity in cancer cell lines, as well as the ability to inhibit early stage colon tumors in rats. In vivo studies our working group has determined that it presents good tolerability, without systemic affectation, however, there were antinutritional effects as the loss in weight gain of 10% in the treated rats. This makes it necessary to study its systemic and residual effects in rats of different ages, considered important in the development of a drug. Healthy Sprague Dawley rats acquired from the INB-UNAM at 5 and 25 weeks of age were used. The treatment schemes were: short-term studies (6 weeks of treatment) in both adult and healthy young rats; intermittent study (2 periods of 6 weeks of treatment and 2 intermediate weeks of rest) with young rats where sacrifices were made on days 1, 9, 22, 43, 57, and 100 to observe residual and systemic effects; sustained study (12 weeks of treatment) with young rats. Weekly the animals were weighed and mesure, the food consumption was determined and at the end of the corresponding treatment, biochemical tests were performed, such as blood biometrics, total proteins, albumin, creatinine, urea, α-amylase, insulin, ALT and glucose. The liver, pancreas, small and large intestine, thymus and spleen were weighed and measured, performing histopathological analysis. No systemic and antinutritional effects were found after subchronic administration of TBFL with adult and young rats, although pancreatic hypertrophy was observed. In the intermittent experiment in young rats there was a decrease in weight gain of 4%, on day 9 there was an increase in the proportion of granulocytes and an increase in the white pulp in the spleen, on day 43 an increase in the size of the small intestine was observed 14% and 21% bowel reduction, intestinal atrophy, as well as pancreatic hypertrophy. After the 2-week rest, pancreatic hypertrophy reverted and atrophy was observed, and by day 100 it was found that the pancreas recovered. In the chronic treatment, there were no systemic or antinutritional effects, except for the increase in heart size. The results show that the administration of TBFL causes adverse effects related to its antinutritional properties without detrimental effect at the systemic level. It will be necessary to determine if the adverse effects are reversed completely using longer resting patterns.
