dc.descriptionListeriosis es una enfermedad con un alta tasa de letalidad que va del 25-50% y es provocada por Listeria monocytogenes, bacteria fuertemente asociada al queso panela. Una alternativa para mitigar este problema es el uso de antimicrobianos naturales, por lo que en este trabajo se evaluaron la endolisina PlyP100 (PlyP100), nisina (NIS), arginato laurico (LAE) y Ɛ-polilisina (EPL) contra el desarrollo del patógeno en queso panela. Se utilizó un coctel de 5 cepas de L. monocytogenes y se evaluó la concentración mínima inhibitoria (CMI) en caldo de cultivo así como el efecto sinérgico, aditivo o antagónico mediante una prueba de tablero. Posteriormente, el queso fue elaborado en un modelo miniaturizado de laboratorio (MLQF), inoculado con 4 log UFC/g del coctel, adicionado con los antimicrobianos solos y en combinaciones dobles a sus concentraciones máximas permitidas por la FDA y almacenado a 4°C durante 28 días. El CMI fue 3.12-6.25 µg/mL, 12.5 µg/mL y 62.50-125 µg/mL para NIS, LAE and EPL respectivamente. En el queso, el tratamiento con PlyP100 fue el más efectivo inhibiendo el crecimiento de la bacteria en 3.50 ± 0.38 log, EPL provocó una inhibición de 1.60 ± 0.67 log, LAE de 1.24 ± 0.06 log y NIS de 0.38 ± 0.44 log en comparación con el control, el cual alcanzó 9.14 ± 0.19 log UFC/g. Sin embargo, la combinación entre EPL y LAE inhibió el crecimiento en 3.50 ± 0.35 log, al igual que PlyP100, incluso utilizando una concentración más baja de LAE en la mezcla. Se encontró un efecto sinérgico entre LAE y EPL tanto en caldo de cultivo como también en la matriz alimentaria. Estos compuestos, EPL y LAE, son generalmente reconocidos como seguros (GRAS) lo cual favorece el uso de esta mezcla para inhibir el crecimiento de L. monocytogenes en queso panela.es_ES
dc.descriptionListeriosis has one of the highest lethality rates among foodborne pathogens; it remains approximately 25-50%. The involved bacteria in this important disease is Listeria monocytogenes which is strongly associated with panela cheese. An alternative to reduce this problem has been the use of natural antimicrobial, therefore in this work, mixtures from phage endolysin PlyP100 (PlyP100), nisin (NIS), lauric arginate (LAE) and Ɛ-polylysine (EPL) were evaluated against the pathogen bacteria in panela cheese. It was used a cocktail of 5 strains of L. monocytogenes determining the minimum inhibitory concentration (MIC) and the synergistic, additive or antagonist effect by a checkerboard test. Later, cheese manufactured in a miniaturized of laboratory model Queso Fresco production (MLQF), was inoculated with 4 log CFU/g of the cocktail, added with antimicrobial compounds alone and in double combinations as the maximum permissible concentration by FDA and then storage at 4°C during 28 days. The MIC was 3.12-6.25 µg/mL, 12.5 µg/mL and 62.50-125 µg/mL for NIS, LAE and EPL respectively. In cheese, treatment with endolysin was the most effective inhibiting Listeria growth in 3.50 ± 0.38 log CFU/g, EPL inhibited the count in 1.60 ± 0.67 log, LAE 1.24 ± 0.06 and NIS only 0.38 log CFU/g compared with the control that achieved 9.14 ± 0.19 log CFU/g. Nevertheless, combination between EPL and LAE inhibited the growth 3.50 ± 0.35 log, as PlyP100, even at using lower concentrations than the maximum permissible of LAE in the mixture. Synergistic effect was found between EPL and LAE in broth culture as food matrix. Those compounds, EPL and LAE, are generally recognized as safe (GRAS) level which favors the use of this mixture to order decreased the L. monocytogenes growth in panela cheese.es_ES
