Description: El jitomate es una de las hortalizas más producidas a nivel mundial y ha sido implicado en brotes de Salmonella spp. y Listeria monocytogenes. El jitomate cherry se consume principalmente crudo y el único tratamiento antimicrobiano que se aplica suele ser la desinfección química. El objetivo de este trabajo fue evaluar el comportamiento de Salmonella spp. y Listeria monocytogenes adheridas a jitomate cherry y su tolerancia a diversos desinfectantes. Se utilizaron cepas de Salmonella spp. y L. monocytogenes resistentes a rifampicina; previo a su uso en estudios de desinfección y comportamiento en jitomate cherry se valoraron diferencias en el desarrollo de cada cepa en caldo soya tripticasa (CST) diluido (0.1, 1 y 10 %), con pH modificado (4, 5 y 6) y con adición de NaCl (1, 1.5 y 2%). También se comparó la susceptibilidad de las cepas resistentes y no resistentes a rifampicina a: NaOCl (30 ppm), agua electrolizada (EW) 30 ppm y ácido acético comercial (2 % para Salmonella y 4 % para Listeria monocytogenes). Se almacenaron jitomates inoculados con ambos patógenos en 4 (7 días) y 22 ºC (5 días). Se cuantificaron periódicamente las células débilmente adheridas (CDA) y las células fuertemente adheridas (CFA) a la superficie del fruto, y se evaluó la eficiencia de la desinfección con NaOCl (60 ppm) y EW (60 ppm) después del almacenamiento. No se encontraron diferencias (α≤0.05) entre cepas resistentes y no resistentes a rifampicina en las diferentes condiciones de desarrollo, ni en la susceptibilidad a desinfectantes. Salmonella y L. monocytogenes fueron capaces de adherirse a la superficie del jitomate. La tendencia de ambos patógenos adheridos a jitomate fue a sobrevivir independientemente de la temperatura; el mayor desarrollo se observó a 22 °C con Listeria monocytogenes (~1 Log UFC/jitomate de incremento). La mayor proporción de CFA se encontró en los jitomates inoculados con L. monocytogenes y almacenados a 4 °C (~3 Log UFC/jitomate) La suceptibilidad a los desinfectantes de Salmonella inoculada en el fruto estuvo influenciada por la interacción entre la intensidad de adhesión (CDA y CFA) y la temperatura, la mayor susceptibilidad se observó en las CDA almacenadas a 22 ºC (~3.24 Log UFC/jitomate); mientras que para L. monocytogenes, el factor relevante fue el tipo de población, Las células fuertemente adheridas fueron las más tolerantes a los desinfectantes. La adhesión de L. monocytogenes y Salmonella al jitomate cherry favorece su sobrevivencia ante los desinfectantes; por ello la desinfección no es suficiente para asegurar la inocuidad del fruto y es necesario minimizar la exposición del fruto a fuentes de contaminación a lo largo de la producción, comercialización y consumo.
The tomato is one of the vegetables more produced worldwide however it also has been implicated in outbreaks of Salmonella and Listeria monocytogenes. Cherry tomato is specially eaten raw and the only antimicrobial treatment prior to consumption is usually chemical disinfection. The aim of this work was to evaluate the behaviour of Salmonella spp. and Listeria monocytogenes cells adhered to cherry tomato and their tolerance to diverse disinfectants. Rifampicin-resistant strains were used, previously, their behaviour between resistant and parental strains were compared. The evaluation of the growth of each strain were made in tripticase soy broth (TSB) diluted (0.1, 1 and 10%), with modified pH (4, 5 and 6) and with the adition of NaCl (1, 1.5 and 2%). Also the susceptibility to disinfectants was tested: NaOCl (30 ppm), electrolyzed neutral water (EW 30 ppm) and comercial acetic acid (2% for Salmonella and 4% for L. monocytogenes). The inoculated tomatoes were stored at 4 (7 days) and 22 °C (5 days). Both, the weakly adhered (CDA) and the strongly adhered cells (CFA) to the surface of the tomatoes were counted after the storage. They were also disinfected with NaOCl (60 ppm) and EW (60 ppm). No differences were found (α <0.05) between the resistant and parental strains in the diferent growing conditions or in the susceptibility to disinfectants. Salmonella and L. monocytogenes were capable to adhere and survive on the tomato surface at 4°C for 7 days. No mater wich storage temperature was used, the trend of both adhered microorganisms to tomatoes was to survive; the greatest increment was observed with L. monocytogenes at 22°C (~1 Log CFU/tomato). The biggest rate of CFA was found in the tomatoes inoculated with L. monocytogenes and stored at 4°C (~3 Log UFC/tomato). The susceptibility of Salmonella to disinfectants was influenced by the interaction between the type of population (CDA and CFA) and temperature, the CDA stored at 22°C were more susceptible (~3.24 Log UFC/jitomate). For L. monocytogenes, the most important factor was the type of population, the CFA were more tolerant to disinfectants than CDA. The attachment of L. monocytogenes and Salmonella to cherry tomato improve the tolerance to disinfectants; therefore disinfection does not guaranty the produce safety and it is necessary to minimize the exposition of the tomato to sources of contamination through production, comercialization and consumption.
