dc.descriptionAntecedentes: el trastorno depresivo mayor (TDM) es la enfermedad psiquiátrica más común. En México, tiene una prevalencia del 12.8% en población adulta de centros urbanos. Se considera un desorden multifactorial y su etiología se asocia a factores ambientales, genéticos y alteraciones fisiológicas, Al respecto, una de las hipótesis postuladas sugiere una desregulación del eje hipotálamo-hipófisis-tiroides (HHT). A sí mismo, existe evidencia que sugiere que el TDM puede presentar una predisposición génica. Hasta ahora se reporta que los polimorfismos de nucleótido sencillo (SNP) rs225014 en el gen DIO2 y los SNP rs10770704 y rs10444412 en el gen OATP1C1, están asociados con la presencia de TDM en pacientes hipotiroideos. A la fecha no existen marcadores biológicos o bioquímicos que permitan mejorar el tratamiento para pacientes con TDM. Objetivo: evaluar la posible asociación en pacientes eutiroideos con TDM y la presencia de los SNP rs225014, rs10770704 y rs10444412. Material y métodos: se evaluaron a 78 sujetos adultos residentes del estado de Querétaro sin antecedentes de patología tiroidea. Se valoró el diagnóstico del TDM por medio del MINI Plus. Se obtuvo una muestra de sangre para genotipificación (qPCR) y para medir HT en sangre (ELISA). Resultados: Las concentraciones séricas medias fT4 (p= 0.0001) y TSH (p= 0.0004) fueron menores en el grupo con TDM. Como era de esperar, la correlación entre fT4 y TSH en los controles fue positiva y significativa (p= 0.0003). Sin embargo, no hubo correlación entre las dos hormonas en el grupo con TDM (p= 0.1061). El grupo con TDM presenta niveles bajos normales de TSH con la presencia de los SNP rs225014, rs10770704 y rs10444412 como diplotipos (p= 0.04) Conclusiones: los pacientes con TDM muestran bajas concentración de fT4 y TSH, sin embargo, los rangos se encuentran dentro de los valores normales. A su vez, los pacientes con TDM muestran una falta de correlación en el eje HHT. Además, el grupo con TDM presenta niveles bajos normales de TSH y la presencia de los SNP. Probablemente, estas diferencias hormonales y la presencia de los SNP podrían llevar a una baja disponibilidad de HT dentro del sistema nervioso central. Esta situación a su vez, podría predisponer al desarrollo TDM.es_ES
dc.descriptionBackground: major depressive disorder (MDD) is the most common psychiatric diseades. In Mexico, it has a prevalence of 12.8% in the adult population. It is considered a multifactorial disorder and may be influenced by environmental factors and physiological alterations. One of the postulated hypothesis suggests a deregulation of the hypothalamic-pituitary-thyroid axis (HPT). There is evidence that suggests that MDD may have a genetic predisposition. So far, it has been reported that single nucleotide polymorphisms (SNPs) rs225014 in the DIO2 gene and SNPs rs10770704 and rs10444412 in the OATP1C1 gene have been associated with the presence of MDD in hypothyroid patients. Those SNPs do not influence circulating serum levels of thyroid hormones (TH). Currently, there are no biological or biochemical markers to improve treatment for patients with MDD. Objective: evaluate the possible association with MDD and the presence of the SNPs rs225014, rs10770704 y rs10444412 in euthyroid patients. Material and methods: the participants are adults residents of the state of Queretaro with no history of thyroidal pathology. The diagnosis of MDD was made by the MINI Plus. A blood sample was obtained for genotyping (qPCR) and measurement of TH in blood (ELISA). Results: Mean serum fT4 (p = 0.0001) and TSH (p = 0.0004) concentrations were lower in the MDD group. As expected, the correlation between fT4 and TSH in the controls was positive and signficant (p= 0.0003). However, there was no correlation between these two hormones for the MDD group (p= 0.1061) The group with TDM presents normal low levels of TSH with the presence of SNP rs225014, rs10770704 and rs10444412 as diplotypes (p = 0.04). Conclusions: patients with MDD show a lower concentration of fT4 and TSH, however, the range is within normal values. Likewise, patients with MDD show a lack of correlation in the HPT axis. In addition, the group with MDD presents normal low levels of TSH and the presence of SNPs. Probably, these hormonal differences ando the presence of SNPs might lead to a low availability of TH within the central nervous system. This situation in turn, could predispose to the development of MDD.es_ES
