Descripción : Introducción: La pegilación comúnmente se realiza de forma química aumentando el peso y tamaño de la molécula con lo que evita la ultrafiltración renal. Además, se caracteriza por presentar una gran velocidad de absorción. En el mercado existen medicamentos que han sido modificados químicamente con Polietilenglicol (PEG), la modificación de estos fármacos han presentado una incremento en la semivida, lo que se traduce a una disminución en la dosis para alcanzar el efecto terapéutico. Dentro de las modificaciones que existen a polímeros hay un método físico donde utiliza reacciones ionizantes con el fin de modificar estructuras poliméricas formando reticulaciones, es decir, al transferir energía inducida por rayos x o gama el polímero abre sus cadenas permitiendo la unión hacia otras cadenas de polímeros para poder formar redes, de tal manera que hay un incremento en su estructura y existe un cambio físico-químico en el material irradiado. Objetivo: Comparar por Dispersión de luz e IR el desarrollo de un método físico de pegilación al cisplatino con la modificación Química del cisplatino utilizando PEG Metodología: Se realizó una modificación Química y Física a tres diferentes concentraciones de cisplatino con PEG de peso molecular 400 Da dando como resultado un mayor peso molecular y tamaño de partícula del medicamento, llevando a cabo pruebas de caracterización por Dispersión de Luz Dinámica (DLS) e Infrarrojo (IR). Resultados: El espectro de IR muestra los grupos funcionales característicos del acoplamiento fármaco-polímero sin alterar su sitio activo del Cisplatino y un incremento de tamaño de partícula por ambos métodos de Pegilación caracterizados por DLS, dando un mejor rendimiento utilizando el método de pegilación de forma física. Conclusiones: Al ser pegilado el Cisplatino por ambos métodos de pegilación aumenta el tamaño de partícula en comparación con el cisplatino sin pegilar. Al comparar el pegilado tradicional con el método físico se observó que existe un mayor número de moléculas pegiladas. El PEG es escindible lo cual no afectara el incremento de tamaño y tampoco su sitio de unión fármaco-PEG para realizar su acción terapéutica ya que se cuidó que fuera en sitios específicos esta modificación en la molécula de cisplatino.
Introduction: Pegylation is commonly carried out chemically increasing the weight and size of the molecule, thus avoiding renal ultrafiltration. In addition, it is characterized by a high absorption speed. In the market there are drugs that have been chemically modified with polyethylene glycol (PEG), the modification of these drugs have shown an increase in the half-life, which translates into a decrease in the dose to achieve the therapeutic effect. Within the modifications that exist to polymers there is a physical method where ionizing reactions are used to modify polymeric structures forming reticulations, that is to say, when transferring energy induced by x-rays or gamma, the polymer opens its chains allowing the union towards other polymer chains to be able to form networks, in such a way that there is an increase in its structure and there is a physical-chemical change in the irradiated material. Objective: Compare by light scattering and IR the development of a physical method of pegylation to cisplatin with the chemical modification of cisplatin using PEG Methodology: A chemical and physical modification was made to three different concentrations of cisplatin with PEG of molecular weight 400 Da giving as a result, a greater molecular weight and particle size of the drug, carrying out characterization tests by Dynamic Light Dispersion (DLS) and Infrared (IR). Results: The IR spectrum shows the functional groups characteristic of the drug-polymer coupling without altering its active site of Cisplatin and an increase in particle size by both PEGylation methods characterized by DLS, giving a better performance using the PEGylation method. physical. Conclusions: Cisplatin pegylated by both pegylation methods increases the particle size compared to non-pegylated cisplatin. When comparing the traditional pegilado with the physical method it was observed that a greater number of pegylated molecules exists. The PEG is cleavable which will not affect the increase in size and neither its drug-PEG binding site to perform its therapeutic action since it was taken care that this modification in the cisplatin molecule was at specific sites.
