Description: Los marcadores genéticos son secuencias que contribuyen a la aparición de una característica de interés, se destaca su importancia para identificar rasgos que no se pueden seleccionar de manera tradicional, tal como la conducta. El comportamiento en la acuicultura tiene un efecto sobre la domesticación y el bienestar del cultivo, destacando la importancia de la conducta de agresividad, ya que afecta directamente la productividad. Para realizar la investigación de marcadores genéticos conductuales se requiere el uso de organismos modelo, y un candidato para el estudio de la agresividad es el pez modelo Astyanax mexicanus. A. mexicanus presenta poblaciones de superficie (PS) y de cueva (PC) que evolucionaron independientemente, obteniendo fenotipos de PS altamente agresivos, con interacciones jerárquicas complejas, y fenotipos de PC que perdieron su agresividad, lo que permite realizar estudios comparativos de alta y baja agresividad. Con el fin de cultivar las diferentes poblaciones de A. mexicanus se diseñó un sistema de cultivo modular con la capacidad de inducir la reproducción de la especie mediante un gradiente de temperatura. Se capturaron PS y PC para su mantenimiento y uso como modelos de estudio, caracterizándolos por su nivel de agresividad. Se estableció una metodología para la cuantificación del nivel de agresividad de la especie mediante el número de ataques por minuto, con una diferencia de más del 60% entre los PS más agresivos y los PC, identificando PS con agresividad alta, PS con agresividad media y PC con muy baja agresividad. Dependiendo de su nivel de agresividad las poblaciones se caracterización mediante la expresión diferencial de genes candidatos al control de la agresividad. Se encontró un efecto en el control de la agresividad debido al transportador de serotonina y al receptor de serotonina B, con una correlación de -0.9 y -0.85 respectivamente. El aumento en la expresión de estos genes se relaciona con la disminución de la agresividad por lo que se propone el uso de la expresión de estos genes como un marcador genético para la identificación temprana del nivel de agresividad en A. mexicanus.
Genetic markers are sequences that contribute to the appearance of a characteristic of interest, highlighting its importance to identify traits that cannot be selected in a traditional way, such as behavior. Behavior in aquaculture influences the domestication and welfare of the crop, highlighting the importance of aggressive behavior, as it directly affects productivity. To carry out the investigation of behavioral genetic markers, the use of model organisms is required and a candidate model for the study of aggression is the fish Astyanax mexicanus. A. mexicanus presents surface (PS) and cave (PC) populations that evolved independently, obtaining highly aggressive PS phenotypes, with complex hierarchical interactions, and PC phenotypes that lost their aggressiveness, what allows comparative studies of high and low aggressiveness. In order to cultivate the different populations of A. mexicanus, a modular culture system was designed with the capacity to induce the reproduction of the species by means of a temperature gradient. PS and PC were captured for their maintenance and use as study models, characterizing them by their level of aggressive behavior. A methodology was established to quantify the level of aggressiveness of the species by means of the number of attacks per minute, with a difference of more than 60% between the most aggressive PS and the PC, identifying PS with high aggressiveness, PS with medium aggressiveness and PC with very low aggressiveness. Depending on their level of aggressiveness, populations were characterized by the differential expression of candidate genes for the control of aggressiveness. An effect was found in the control of aggressiveness due to the serotonin transporter and the serotonin B receptor, with a correlation of -0.9 and -0.85 respectively. The increase in the expression of these genes is related to the decrease in aggressiveness, therefore the use of the expression of these genes as a genetic marker for the early identification of the level of aggressiveness in A. mexicanus is proposed.
